Researchers May Have Found Equivalent of Embryonic Stem Cells

Chinese scientists have bred mice from cells that might offer an alternative to human embryonic stem cells, producing the most definitive evidence yet that the technique could help sidestep many of the explosive ethical issues engulfing the controversial field but raising alarm that the advance could lead to human cloning and designer babies.

In papers published online Thursday by two scientific journals, separate teams of researchers from Beijing and Shanghai reported that they had for the first time created virtual genetic duplicates of mice using skin cells from adult animals that had been coaxed into the equivalent of embryonic stem cells.

The findings were welcomed by supporters and opponents of human embryonic stem cell research as a long-sought vital step in proving that the cells could be as useful as embryonic cells for studying and curing many illnesses.

The results come just as the Obama administration has eased federal restrictions on government funding for embryonic stem cell research, and they could influence how to prioritize millions of dollars in new spending in the field (…)

“The implications of this are both enormously important and troublesome,” said Robert Lanza, a stem cell researcher at Advanced Cell Technology in Worcester, Mass. “It revives many of the issues raised by reproductive cloning.”

Many scientists believe human embryonic stem cell research could revolutionize medicine by enabling doctors to use genetically matched tissue to treat many diseases. But the field has been mired in controversy because embryos are destroyed to obtain the cells.

In 2006, scientists discovered that they could induce adult cells to regress to a stage that appeared identical to embryonic stem cells, called induced pluripotent stem (iPS) cells. Although scientists have become increasingly adept at creating and manipulating such cells, questions have lingered about whether they are truly equivalent. The new experiments were designed to put the cells to what has long been considered the most rigorous test.

In the studies, published in the journals Nature and Cell Stem Cell, the researchers used viruses to flip genetic switches in the DNA of skin cells from adult mice to turn them into iPS cells in the laboratory. The researchers then injected some of the iPS cells into very early embryos that are capable of forming a placenta but not of fully developing on their own. The resulting embryos were then transferred into the wombs of surrogate mice (…)

The second group of researchers, led by Shaorong Gao of the National Institute of Biological Sciences in Beijing, created five iPS cell lines, one of which was able to produce embryos that survived until birth. Although four animals were born, only one lived to adulthood. Nevertheless, the work is “proof that iPS cells are functionally equivalent to embryonic stem cells,” Gao said in a telephone interview.

Other researchers agreed, praising the work as a long-awaited confirmation of the cells’ equivalence.

“This clearly says for the first time that iPS cells pass the most stringent test,” said Konrad Hochedlinger, a stem cell researcher at Harvard University.

Opponents of human embryonic stem cell research said the findings provide the latest in a growing body of evidence for why such research is no longer necessary.

“Nobody has been able to find anything that embryonic stem cells can do that these cells can’t do,” said Richard M. Doerflinger of the U.S. Conference of Catholic Bishops. “This was the last remaining barrier.”

The Chinese scientists and others, however, said continued research on embryonic stem cells remains crucial to validate iPS cells and because it remains unclear which cells will turn out to be most useful for different purposes.

But the cells’ ability to produce almost genetically identical offspring raised the fear that rogue scientists might misuse the technique to attempt to clone humans.

“The culture wars are not over,” said Jonathan D. Moreno, a University of Pennsylvania bioethicist. “There was a lot of celebration about the end of the ethical issues with induced pluripotent stem cells. But this is the paradigm case that shows that the old debates are rapidly being transformed into something even more complicated.” (…)

Fox, an internationally renown star since “Back to the Future”, told weekly magazine ‘Grazia’ how he discovered his illness and what he has experienced in the past years.

At the age of 30, his career was cut short by Parkinson’s disease. Now, after years of silence, isolation, alcohol, and crisis (“Initially I reacted with rage, I just wanted to drink so I could avoid dealing with the situation,” said the actor), thanks to the help of his wife, Tracy Pollan, he has written a book: “Lucky Man” and has returned to television with “Michael J. Fox: Adventures of an Incurable Optimist”, a trip around the world interviewing well-known people, and not only about their idea of optimism. “I am investigating into the nature of optimism and hope, and the level of acceptance that helps you persevere and face every day in the best way possible.

Those who are sick will never know how nice it is to feel well again.” He says that he no longer wants to return to film-making: “I will not make films anymore. I don’t miss anything about the film industry. The ‘Stuart Little’ films (he was the voice of the mouse, editor’s note) were ideal for me because I just had to sit there in a dark room with headphones and a microphone”.

Now involved in politics and stem cell regulation reform, he finances research into Parkinson’s through his association, “The Michael J. Fox Foundation”. The only advice that he wants to give to people is to never forget that every day, in every moment it is possible to invent the future that you want. Death is the ending of everyone’s story. We all have to die sooner or later. Once you accept this, the problem becomes the quality of your life.”

Experts dispute doctor’s stem cell breakthrough claim

SANTIAGO, Dominican Republic (CNN) — This Caribbean city already known for cigars, furniture, chocolate and coffee may become a magnet for Americans seeking controversial stem cell therapy for life-threatening illnesses if a Florida cardiologist has his way.

The Food and Drug Administration has not approved this stem cell therapy in the United States because no clinical trials to prove its effectiveness have been done. But Dr. Zannos Grekos says his company, Regenocyte Therapeutic, has successfully used adult stem cells to treat patients with heart and lung disease.

Grekos said he and his associates draw blood from a patient in Florida and then send it to a laboratory in Israel that produces what his company calls “regenocytes.” The company defines regenocyte as “a stem cell that has been activated to become a target organ.” (…)

But Grekos’ procedures have not been reviewed by other researchers, and leading scientists involved in U.S. stem cell research efforts say Grekos is simply wrong. Dr. Irving Weissman, president-elect of the International Society for Stem Cell Research, told CNN, “There is no such cell. There is nothing called a ‘regenocyte.’ ”

“As a stem cell scientist who works in the field of regenerative stem cells, I am disappointed and shocked that somebody would prey on a family that has an untreatable disease with the promise of a therapy that has no scientific or medical basis,” Weissman said.

Grekos has a busy practice in Bonita Springs, Florida, outside Naples, and runs a company that promotes and administers stem cell therapies in Santiago, a noisy, crowded industrial city in the central Dominican Republic.

He told CNN that in the past 18 months, about 100 patients have received adult stem cell therapy at a Dominican hospital. Most of them have been patients with severe heart disease, while the rest have suffered from chronic lung illnesses, he said.

He said the Israeli laboratories can produce between 40 million and 80 million stem cells from a patient’s blood sample (…)

The procedure is costly. Grekos said he charges $64,500 — none of it covered by insurance. He said the cost is driven by the expense of processing the stem cells, and providing clean rooms and couriers who hand-carry the cells to Israel and back.

A year ago, Barbara McKean of Naples, Florida, paid Regenocyte Therapeutic more than $54,000 for stem cell therapy in the Dominican Republic after suffering from chronic lung disease. She said she believes the therapy was worth the expense (…)

The chief medical officer of the American Lung Association, Dr. Norman Edelman, said he does not doubt the sincerity of patients who believe they have been helped by stem cell therapy. But he added, “There’s an enormous placebo effect in almost all of these cases.” (…)

Stem cell experts who reject his methods “need to be better educated,” he said.

ITALY – Gitmo: Italy one of the top ranked counties in Europe for bone marrow stem cell transplants

Italy is ranked among the top countries in Europe for the number of bone marrow transplants performed. This is one of the elements that emerged on the first day of the 3rd GITMO conference (Italian group for bone marrow transplants), which began today in Florence.

Italy plays a leading role for bone marrow transplants, said a statement, demonstrated by the fact that our country is one of the top contributors to the number of transplants performed in Europe. The European Bone Marrow Transplant Group (EMBT) collects and stores data regarding transplants performed in various countries, and Italy is ranked 3rd/4th place for the total number of transplants it performs. This is an indication of the important work performed in Italy by GITMO and demonstrates that Italian transplant centers make use of top-level expertise and technology.

This is encouraging data because it opens up possibilities for the future, with important implications for patients suffering from neoplastic diseases, for which stem cells could play a key role in future treatments.

The procedure of stem cell transplants is very invasive for the patient and not all individuals are able to adequately tolerate the procedure, especially older patients. In the past, it was not possible to perform a transplant in an individual over the age of 40, then the age was raised to 50, while today 60 could be an important objective. In the past 10 years, transplant technologies for family and non-family donors have been developed with the use of so-called reduced intensity transplants, meaning a transplant that uses technology to make the procedure tolerable for patients who are not able to undergo the traditional procedure.

Initially, this technique developed slowly, while today many transplant centers have adopted the procedure, which has substantially increased the number of people who are eligible to receive this type of treatment. For some diseases, like acute myeloid leukemia, donor stem cell transplants represent our most promising weapon, and since these diseases are most common in elderly individuals, in cases where the patient is in good condition and they are under 60 years old, it is possible to use this type of treatment.

Future prospects. Today stem cells can be obtained from various sources. While in the past family donors were used, now the use of non-family donors has become more widespread thanks to international donor banks. Furthermore, alternative sources are available, like umbilical cord stem cells, which have been collected in many EU and non-EU countries for about 15 years in special umbilical cord blood banks. They are also present in Italy and this expands the offer and possibility to quickly find a source of stem cells to be used in transplants.

ITALY – Umbilical cord stem cell collection starts with info in pharmacies

Getting information and advice in the pharmacy to collect and store umbilical cord stem cells abroad: this is the service provided by the Clic-care system, a brand managed by a company operating in the home assistance sector, and already present in pharmacies in Milan. The company recently issued a statement presenting the service. A service in which each pregnant woman, according to the company’s website www.clic-care.it, can request directly at the pharmacy to reserve a free consultation appointment for information on storing umbilical cord stem cells. Storage which, according to Italian law, can only be done in public stem cell banks for public use, not in private banks.

After a free telephone interview in which women can learn and understand the procedure to store stem cells, the company promises that it will take care of all of the paperwork required by Italian law as well as practical aspects of the process. After birth, the cells are handed over to specialized technicians in the hospital who will send them to the United Kingdom, where the cells will be deposited and stored in a private center. Periodically, the family will be informed about new goals and available cures. The president of the Italian National Federation of Pharmacy Owners (Federfarma), Anna Rosa Racca, said, “currently, Federfarma is collecting information on the initiative to assess the various aspect”.

Exporting umbilical cords abroad to be stored and collecting umbilical cord stem cells for autologous use “is possible if done according to Italian law. It is necessary to evaluate whether the other services offered by private companies in Italy respect all of the laws,” said Alessandro Nanni Costa, the director of the National Transplant Center (CNT) said regarding clic-care.

In Italy, said Nanni Costa, “collecting stem cells for autologous use and private banks to store them is not allowed. For exports abroad, we offer counseling to inform pregnant women about collection methods, storage, and use. The women must fill out a form about autologous collection on the CNT website”. Then they must obtain the necessary certification from the regional Health Department, and then must ask for authorization from the Health Ministry.

When this procedure has concluded, the umbilical cord can be sent abroad. “If all of these regulations are satisfied, along with all of the other requirements, like those for health-related advertisements, the service offered by these private companies is legal. But it is necessary to assess whether the information is correct and to understand exactly what they offer and how. If the information is not correct, then we will evaluate the timetable and methods to intervene”.

CNN Headline Story Focuses on Stem Cell Research Doctor

In the Dominican Republic, Dr. Zannos Grekos, a stem cell research doctor, is now helping patients with heart disease and lung diseases with their own Adult Stem Cells as reported on CNN in a special research report.

Adult Stem Cells Helping Heart Disease and Lung Disease

In the last 18 months, Dr. Grekos and his company Regenocyte have treated more than 100 patients for either heart diseases such as congestive heart failure, coronary artery disease, heart attacks, and angina and lung diseases such as pulmonary hypertension, emphysema and COPD.

The story features Barbara McKean of Naples, Florida. Barbara was suffering from the lung disease, COPD and was treated with her own stem cells.

His Eyes Are Wide Shut

Check out this quote from the chief medical officer of the American Lung Association-

The chief medical officer of the American Lung Association, Dr. Norman Edelman, said he does not doubt the sincerity of patients who believe they have been helped by stem cell therapy. But he added, “There’s an enormous placebo effect in almost all of these cases.”

So Dr. Edelman sees a woman, Barbara McKean, featured previously here,  who was on oxygen 24/7 walking around with a cannister and now, after stem cell treatment, is completely off oxygen and he thinks it is the placebo effect? Ridiculous.

Luckily, Barbara’s doctor does not intentionally close his eyes to the obvious:

McKean’s family physician, Dr. Robert Folsom, confirmed to CNN that she had been suffering for many years from an advanced state of COPD, an incurable lung disease. Folsom told CNN he was “quite incredulous” after seeing her condition improve.

Folsom, who said he has been McKean’s family physician for many years, disputed any suggestion that her apparent recovery stemmed from a “placebo effect” — an improvement sometimes seen in patients who are given an inert substance in clinical trials.

“I know about the placebo effect, and her improvement does not seem to be a result of that,” he said.

Stem Cell Therapy- Documenting Results

Dr. Grekos is documenting research data  from his patients treated with their own stem cells and wishes to present it to the FDA at the end of this year.

In the past few days, another stem cell research doctor, Dr. Roberto Fernandez Vina, also just presented his results of his stem cell research data for COPD and Emphysema at the annual International Society for Stem Cell Research annual meeting.

And Dr. Grekos had a parting shot for the “experts” who always try to downplay any adult stem cell success story:

He shrugs off the criticism from people who doubt his claims, saying he — and not they — are on medicine’s cutting edge.

Stem cell experts who reject his methods “need to be better educated,” he said.
